Raman Spectroscopic Analysis of Drug Delivery Systems

ABSTRACT

This presentation summarizes the utility of Raman spectroscopy for characterization of various structural aspects of drug delivery systems in order to improve their performance. Particular emphasis is given to drug eluting stents. The examples cited illustrate drug distribution in the overall structure, miscibility behavior of various polymers used and dynamics of polymers which control the release mechanism. Release kinetics in many drug delivery systems have been established to be dependent on phase separated structures of multi-component systems involving various polymers and low molecular weight drugs. High resolution Raman microscopy has proven invaluable in following the kinetics of structural evolution, attained local chemical composition and their changes as a function of time and temperature. By use of the high resolution Raman technique, the manufacturing process of various biodegradable polymeric stents can be characterized even at the micron level. Such directly obtained structural information contributes to better design of various drug delivery systems.
